Drug Name
Clofibrate (Atromid-S) --
Mechanism of action is unknown. Lowers serum triglycerides and very low-density lipoprotein levels. Serum cholesterol and low-density lipoprotein levels are lowered less predictably and less effectively.
Adult Dose 500 mg PO qid
Pediatric Dose Not established
Contraindications
Documented hypersensitivity; severe hepatic or renal dysfunction; primary biliary cirrhosis
Interactions
May potentiate effects of warfarin, insulin, and sulfonylureas; coadministration with dantrolene may decrease protein binding of dantrolene; coadministration with ursodiol may counteract effects of ursodiol; oral contraceptives may increase elimination; probenecid may increase effects (may need to decrease clofibrate dose)
Pregnancy C - Safety for use during pregnancy has not been established.
Precautions
Possible increased risk of malignancy and cholelithiasis; monitor for abnormal elevation of ALT, AST, LDH, bilirubin, and alkaline phosphatase serum levels; caution in patients with peptic ulcer disease; cardiac arrhythmias have been reported; muscle aches, soreness, and cramping may occur

Das heißt also man weiß nicht wie die Substanz funktioniert. Das ist denke ich MÖGLICHERWEISE ein gewisses Risiko.

Has anybody tried clofirate? I read a couple of articles that say it lowers bili. I know in the interduction page they say its bad for Gilberts so Im a little confused.

Yes, I have - it works on jaundice pretty well. I'd say I'm 90% cleared up. I don't think it is bad for people with Gilbert's - my Dr. has assured me at least. 600 Mg, twice daily.dmy222

Browsin the web as many gs sufferers do - some sites don't recommend this drug citing the fact that it's poorly metabolised.
However there are medical websites stating how beneficial this drug is for gilberts syndrome

It maybe time to revisit the doctor now that there's a drug with seemingly less side effects than phenobarbitol that appears to clear jaundice.

djk - hope you don't mind if i enquire about your experience with the drug in question?
Did you experience fatigue/ brain fog or any other commonly recognised symptoms (by those whom live with the syndrome) of gilberts syndrome?
If yes has each sympton improved/worsened or has there been no change.
Also how long have you been on this drug (a) in total (b) before recognising improvement.

Sorry for the long period of time before getting back on this site/forum. I've been on Clofibrate so long I can't remember how long it looks for jaundice to get better, maybe a month or so. I remember being amazed and quite happy because I went from easily noticeable jaundice to hard to notice or not noticeable. However, knowing myself, I still can tell. I've noticed that when I work in the sun for a while (gardening, etc.) it also makes my eyes clearer for a while.
I never really experienced the brain fog (or maybe I'm just so used to it I don't know any different). I'm tired in the morning, and I didn't realize any difference between before and after the Clofibrate. Clofibrate is also a cholesterol lowering drug, so if you have heart disease in your family I don't think it would hurt. I had heard that phenobarbitol was too harsh, and this is not supposed to be.
